Step Two of the perimenopause blood test measures progesterone during the luteal phase of your cycle indicating whether or not you have ovulated. While this test is normally taken on day 21 of your cycle (for those with a textbook 28 day cycle) the day in which to take the test will vary depending on your cycle length. The best day to take this test is 7 days prior to the start of your period. If your period is irregular you can test every 7 days until to onset of your period.

Typically, progesterone levels are measured around day 21 of a 28-day menstrual cycle, which corresponds to the peak of progesterone production in a natural menstrual cycle. If you are taking hormonal contraceptives or undergoing hormone replacement therapy, your healthcare provider may provide specific instructions regarding the timing of the test. It is important to follow these instructions to ensure accurate interpretation of your progesterone levels.
